Thank you Sejal for this interesting question.

OOC technology represents a promising and innovative approach for toxicity testing and drug development. One area where OOC has been explored greatly is in drug metabolism to overcome the limitations of traditional in vitro cell culture systems in assessing chronic drug exposure and long-term effects on drug metabolism. For instance, liver-on-a-chip devices have been employed to investigate nonalcoholic fatty liver disease (NAFLD). However, it is important to note that the current physiological relevance of these culture systems is still somewhat limited. While OOC holds promise, it is still an evolving field and challenges remain in terms of achieving full organ complexity, long-term functionality, and scalability.

I am particularly interested in the application of OOC technology to create a blood-brain barrier on a chip (BBB-on-a-chip) and curious to hear from fellow members about their experiences on utility of BBB-on-a-chip and how it has been applied in their projects.

Dr. Mallick, thank you for sharing your thoughts on OOC technology and its potential in toxicity testing and drug development. The application of OOC technology to create a blood-brain barrier on a chip (BBB-on-a-chip) is indeed an intriguing area of research with significant implications.

    Hello Sejal,

You can read the following paper (PMID: 34831406). It is a nice paper on BBB model in the chip.

Dynamic 3D On-Chip BBB Model Design, Development, and Applications in Neurological Diseases - PMC (nih.gov)

    Thank you,

Thank you Abhisheak for this nice review article.

Has anyone had experience utilizing Microfluidic BBB models to assess small molecule permeability across the blood-brain barrier? I am interested in learning about its relevance and usefulness in early-stage drug development.
