We have been trying to develop a neutralizing antibody assay for ADA develop after infusion of CAR T cells. We took the approach of establishing a cell based assay based on our in vitro cytotoxicity assay of CAR T cells to a target cells line. This same cells based assay is the potency assay for release of the drug product. It been very difficult to actually neutralize the killing of CAR T cells. Has anyone had the same challenge? Has anyone been successful in developing a cell based Nab assay to further characterize ADA against CAR T cells? Any suggestions or protocols would be helpful? Anyone has seen a protocol for Nab assay for CD19 CAR T cells?
