Dear Reseachers with experience in LNP analysis,
Hi! In order to check the distribution of ionizable lipid (cationic lipid) of LNP (Lipid nanoparticle) in blood and tissues, I would like to conduct LC/MS/MS analysis. I'm trying with known cationic lipids such as SM10, using plasma to establish LC/MS/MS conditions. But it's difficult.
Neat solution seemed to be unstable when water was in it, and when plasmal samples were reinjected within 24 hours, the peak area decreased.
Organic solvent for stock solution : methanol or DMSO
Sample preparation : protein precipitation (plasma : Acetonitrile = 1:5), incubation at 4C for 10 minutes and centrifuged.
Mobile phase : water and acetonitril or methanol
Column : Poreshell 2.1x50 mm
Please advise on anything, such as sample preparation, mobile phase condition, and column.
