ISI Template Harmonization

    Hi all -

    We wanted to reach out and follow up on a discussion during our panel re: NAB results reporting harmonization at NBC this year. There was interest around a working group focused on standardization of the Integrated Summary of Immunogenicity (ISI) with a recommended harmonized template. 

    I recognize that there is a separate work stream for Immunogenicity Risk assessments and other areas, this effort could add value and complement the work there as well as from standardization of reporting immunogenicity results. 

    Looking forward to  hearing feedback from this community here and/or please feel free to reach out to me or the other panelists from the NAB discussion at NBC, Darshana Jani, Susan Irvin, and Amanda Hays.

    I would not mind seeing an effort on standardizing where the details of immunogenicity assays appear in submissions in the CTD format.  While it is fairly standardized that PK assays are described in 2.7.1 along with the associated tables described in the FDA guidance in the 2.7.1 appendices, the location of immunogenicity assay details and validation summaries is less defined.  Across various submissions, I have seen this information presented in 2.7.1, 2.7.2, and the ISI; in some cases, it is presented redundantly in all three places.  I think it should be in the ISI, as it would tie into the assessments made with the data the assays generate.  If this were one objective of your ISI standardization effort, count me in.

    Very interested in joining the working team on this proposal. I can bring the relevant folks from JnJ who had many years of experience filing ISI with multiple programs and seen the evolution on clinical immunogenicity reporting.

    The EMA published an outline of ISI contents quite a few years ago now, which the FDA seemed ok with-is that not being used by sponsors?  Is there a need to modify it?  It was kind of new when I retired and started consulting so I have little experience putting one together, thus my questions.

    Has anyone had thoughts about how standardizing or revising reporting templates would work with the FDA's review tool, Elsa?  For example, is there a particular format that the tool was trained on and changing some items would create issues for the AI review portion?
